| Gloucester ran in three tries at Adams Park to end Wasps' record eight-game winning streak, which dated back to their defeat to the Cherry and Whites at Kingsholm in January.
Ryan's men are now on the verge of a home semi-final, which will be confirmed if Wasps fail to pick up maximum points at Newcastle in midweek.
But with Gloucester still coming of age as a side, Ryan has urged them to see off in-form Bath next week and finish the regular season top of the pile.
"It is important. We have a good record of finishing at the top," said Ryan.
"I want to try and maintain that next week but it is more important we get used to winning big games.
"Next week is an enormous game and whoever wins it will be at the top.
"We have double-banked Wasps in the league. They came into the game at Kingsholm on the back of five wins and we beat them. They came into this game on the back of eight wins and we beat them.
"It shows we can compete at this level and we are in the play-offs. We have got to play in these type of games regularly and we will get better.
"We know we are a good side when we have the ball. But just because we won, it is not a guaranteed thing that we have matured.
"We have still got a lot of growing up to do. We still saw cracks today and on another day it might have been exploited."
Gloucester surged into an early 12-0 lead with tries from James Simpson-Daniel and Mike Tindall before Wasps hit back with the first of two tries from Raphael Ibanez.
Gloucester pulled 22-10 clear with a try from Olly Morgan shortly after the interval. Ibanez struck again to set up a grandstand finish but Wasps were eventually denied a losing bonus point by Ryan Lamb's late penalty.
That saw Wasps slip out of the top four but with Newcastle and Leeds to come in the next six days captain Lawrence Dallaglio is convinced they will reach the play-offs.
"We will make the play-offs, make no mistake about that," he said.
"Games like this are a wake-up call for the sort of players we have in this squad."
Wasps were upset by what appeared to be a high tackle on Cipriani in the build-up to Morgan's try.
Head coach Shaun Edwards said: "It looked pretty high to me but the referee is the sole adjudicator and we not here to criticise referees.
"Danny is an up-and-coming star and if he keeps getting slapped around the head he wont be as pretty as he is for long."
